Das Amid-Prodrug (Etodolac-Glycin-Konjugat) wurde durch Kopplung von Etodolac mit Glycin synthetisiert. Das synthetisierte Prodrug wurde durch Schmelzpunkt, D�nnschichtchromatographie, RM-Werte, Rasterelektromikroskopie, R�ntgenbeugungskristallographie und Hochleistungsfl�ssigkeitschromatographie charakterisiert. Die L�slichkeit in Wasser und die Lipophilie (log P) wurden in verschiedenen pH-Medien bestimmt, n�mlich in HCl-Puffer pH 1,2, saurem Phthalatpuffer pH 4,0, Phosphatpuffer pH 6,8 und pH 7,4. Die In-vitro-Reversion des Etodolac-Glycin-Konjugats zu Etodolac wurde bei verschiedenen pH-Werten, einschlie�lich der Dickdarmumgebung, durchgef�hrt. Die In-vitro-Umwandlung zeigte, dass das Prodrug bei allen anderen getesteten pH-Werten intakt blieb, mit Ausnahme des Dickdarm-pH-Werts, bei dem die mikrofloralen Dickdarmenzyme (Amidase) die Amidbindung des Etodolac-Glycin-Konjugats hydrolysierten und das freie Medikament freisetzten.
